征服了约旦河以东各城后（3:8-17），摩西发现有必要在这些城市设立其中三座，给非故意杀人者提供庇护。因此，在申命记4:1-40的训诫后，叙述者告诉我们，摩西在约旦河东，向日出之地，分定三座城，使那素无仇恨、无心杀了人的，可以逃到这三城之中的一座城，就得存活，"等他站在会众面前听审判"（民数记35:12）。
有关选择逃城的指示也出现在申命记19:1-13和民数记35:9-34中。实际上，依据民数记35:9-34（参见约书亚记20:1-9），总共指定了六座城市：三座在约旦河的东岸，三座在约旦河的西岸。因为申命记4:41-43所列的只是约旦河的东岸，这里就不对约旦河西岸的各城市进行评论。
在申命记4:43，我们知道在约旦河东岸的第一座逃城叫比悉。这座城在旷野平原，并特别指定给了流便人。依据约书亚记21:36和历代志上6:78，比悉后来被分定作为利未人的城市，被分配给了米拉利家族。第二座逃城是基列的拉末。它被指定给了迦得人。这座城市可能就是今天的Tell Ramith。第三座城市是巴珊的哥兰，它被指定给了玛拿西人。哥兰后来分定作为利未人的城市，归给了革顺的子孙（约书亚记21:27；历代志上6:56）。这三座城市都被分定给了流便、迦得和玛拿西各支派，以备有人不小心误杀了他人。
